Departs from the terminus at Bay 8 in Southland Shopping Centre and runs via Nepean Highway and Charman Road, then via Cheltenham Railway Station, along Charman Road, Balcombe Road, Tramway Parade, Martin Street, Holding Street, Reserve Road, Pacific Boulevard, Haydens Road, Fourth Street, Ebden Avenue, along Bluff Road then Royal Avenue, Fernhill Road to Sandringham Station. It then runs via Beach Road, South Road, Brighton Beach Station, down New Street, Bay Street, St Kilda Street, Ormond Road, Broadway, Mitford Street, Barkly Street, Grey Street, Fitzroy Street (St Kilda Light-Rail Station).
